Tryptophan is the raw material for serotonin, but the conversion needs vitamin B6 too, which is why the two appear together in Calm and why the dog studies on tryptophan point to a role alongside other ingredients rather than as a standalone fix.

From food to serotonin

Tryptophan has to come from diet, and once it's available, the body converts it into serotonin through a pathway that also needs vitamin B6, which is why the two appear together in Calm rather than tryptophan working alone.

A body running low on tryptophan has less raw material for that conversion. Supplementing it is meant to make sure the pathway is not limited by supply.

What the studies in dogs found

A study of privately owned, mildly anxious dogs looked at dietary tryptophan supplementation specifically, and a more recent 2024 review asked directly whether supplementary tryptophan reduces signs of anxiety in adult dogs. A 2025 trial went a step further, testing treats combining tryptophan with cannabidiol and alpha-casozepine and finding a mild stress-reducing effect.

That last study combined tryptophan with other actives, so it doesn't isolate tryptophan's individual contribution. Taken together with the anxious-dogs study, though, it supports tryptophan's place as one part of a multi-ingredient approach to mood, rather than a single fix.

Does tryptophan make dogs drowsy

Tryptophan's reputation for drowsiness, the idea that turkey makes people sleepy, comes from its role as a building block for serotonin and, further down that pathway, melatonin. It's a reasonable question to ask about a dog chew.

In the dog studies, though, what gets reported at the amounts used isn't sedation. It's a steadier mood and less anxiety-related behaviour. Owners describe a calmer dog, not a drowsy one, which is why tryptophan sits alongside Calm's other non-sedating ingredients rather than functioning as a sleep aid.
